Data are mean cpm ± 1 SD and are representative of three separate experiments. Responders: T-cells (2 × 103) purified from mesenteric lymph nodes of NOD mice 4 weeks after intravenous administration of DCs (2 × 106) that were propagated from bone marrow of NOD mice in the absence or presence of NF-κB ODN (10 μmol/l). T-cells from NOD mice without DC treatment were used as controls. Stimulators: γ-irradiated DCs propagated from NOD mice bone marrow that were pulsed with islet antigens from NOD mice in vitro (Ag+) or not pulsed with antigens (Ag). Responders were cultured for 5 days with stimulator at an stimulator:responder ratio of 1:10. Cytokine levels in the culture supernatants were determined by ELISA.
